What 1,469 Metabolites Taught Us About Mushroom Extracts

When we founded AHARA, we asked a simple question: what's actually in a mushroom extract? Not what the label says. Not what the Certificate of Analysis checks for. Everything. Every compound, every metabolite, every bioactive molecule.

Most mushroom supplements only measure 2-3 markers. We measured everything.

Why Nobody Else Does This

Metabolomic analysis is expensive. A single dual-panel mass spectrometry run costs thousands of dollars and requires specialized laboratory equipment and expertise.

Most mushroom supplement companies rely on Certificates of Analysis (CoA) that check for heavy metals, microbial contamination, and maybe total polysaccharides. That's quality control, not composition analysis. It tells you the product is safe to consume. It tells you nothing about what's actually in it.

Standard testing measures 2-3 marker compounds out of 1,469. That's characterizing 0.3% of the product's bioactive profile.

What We Found

The results reframed our understanding of what a mushroom extract actually is:

  • Adenosine + adenine, verified in Reishi. The same compounds your brain uses to build sleep pressure.
  • 26 ganoderic acid variants, triterpenes that cross the blood-brain barrier and modulate GABA, suppress inflammation, and inhibit histamine release.
  • Oleamide, a fatty acid amide that signals sleep through the endocannabinoid system. We didn't expect to find this.
  • GABA-pathway compounds, precursors and modulators that support GABAergic signaling.
  • Hericenones + erinacines, the NGF-stimulating compounds in Lion's Mane, verified at meaningful concentrations.
  • Short-chain fatty acids, gut-brain axis mediators that support the microbiome.
  • Cholinergic precursors, supporting acetylcholine synthesis for attention and memory.

17 metabolic pathways. Not one compound, one receptor, one mechanism. An entire biological system.

Why Extraction Method Is Everything

Here's the uncomfortable truth for the industry: most of these compounds don't survive conventional extraction.

Hot water extraction captures polysaccharides but destroys volatile terpenes and heat-sensitive compounds. Alcohol extraction captures triterpenes but denatures proteins and degrades enzymatic compounds. The dual extraction method (water + alcohol) is better than either alone but still misses significant portions of the metabolomic profile.

AHARA's water-based extraction process preserves the full spectrum. No alcohol denaturation. No heat destruction of thermolabile compounds. The result is an extract that actually contains what the mushroom contains, not a stripped-down fraction of it.

This is why we ran the metabolomic analysis in the first place. We needed to verify that our extraction process was preserving what we believed it was. The data confirmed it, and revealed compounds (like oleamide) that we hadn't even been looking for.

What This Means for You

When you see a mushroom supplement label that says "500mg Reishi extract, standardized to 30% polysaccharides," you're looking at a product characterized by one metric out of hundreds that matter.

It's like buying a car based solely on the paint color. The engine, transmission, brakes, suspension, none of that is on the label.

We publish our metabolomic data because we believe the industry needs a transparency standard. Not proprietary blend labels. Not meaningless "10:1 concentration" ratios. Actual molecular composition data.

If a company can't tell you what's in their extract beyond polysaccharide percentage, they don't know what they're selling you. And they definitely can't tell you why it works, or doesn't.
